Test principle
L- Aspartic acid+α-Ketoglutaric acid AST Oxaloacetic acid+L-Glutamate
L-Glutamate+ NADH+H+ MDH L-Malic acid+NAD+
In the coupling reaction above, the oxidation rate of NADH is proportional to the activity of AST. The unit activity of AST can be calculated from the decreasing rate of absorbance detected at a wavelength of 340 nm.
